For laboratory analysis, blood (2 ml) was collected by venipuncture of the jugular vein into sodium heparinized tubes. Blood was centrifuged at 2,500 g for 15 min. Plasma was stored at -20°C for 6 months and at -70°C for 3 months until tested. Plasma concentrations of metabolites, enzymes, and electrolytes were analyzed by the automated biochemical analyzer (Humalyzer 3000, USA).
The manual analyzer results were compared with those obtained by the automated analyzer. Metabolite concentrations should be compared on the basis of the same aliquot of the same sample; to evaluate some of the analytes, aliquots from two sites within the same sample were analyzed by two methods and the reference method was used to compare the results obtained by the manual and automated methods. Student’s t-test was used to determine any significant difference between the results obtained by the test method and that by the reference method.
The accuracy of the AST test was evaluated by the method of comparison between the results obtained by the automated method and those by the manual method. The correlation coefficient and percentage recovery between the results were determined.
